Modelling the helicase to understand hepatitis

NS3 behaves like a “caterpillar” and helps the virus to replicate

NS3 is an enzyme specific to the hepatitis C virus. If developed, a drug capable of recognizing and selectively attacking it could fight the disease without side effects for the body. However, to be able to develop one we need to know more about the behaviour of this important protein in the virus replication process. Until now, we have only had some “still images” – obtained with crystallography techniques – of the interaction between the protein and the RNA, which makes the viral genome. Now some SISSA scientists have used numerical techniques to reproduce the whole “film”, thereby providing a detailed and comprehensive view of the behaviour of NS3. The study has been published in the journal Nucleic Acids Research.

NS3 è un enzima specifico del virus dell’epatite C. Un farmaco che fosse in grado di riconoscerlo e attaccarlo in maniera selettiva potrebbe combattere la malattia senza effetti collaterali per l’organismo, ma per progettarlo serve conoscere nel dettaglio il comportamento di questa proteina importante nel processo di replicazione del virus. Finora erano disponibili solo alcuni “fotogrammi” – ottenuti attraverso tecniche di cristallografia - della proteina nel suo processo di interazione con l’RNA, che costituisce il genoma del virus. Ora gli scienziati della SISSA hanno riprodotto con tecniche numeriche l’intero “film”, offrendo cosi un quadro dettagliato e  complessivo del comportamento di NS3. Lo studio è stato pubblicato sulla Rivista Nucleic Acid Research.
